Interactions of Hedgehog and Ras signaling pathways in rhabdomyosarcoma

Rhabdomyosarcoma (RMS) is the most common soft tissue sarcoma in children. Recently, we identified aberrant Hedgehog (Hh) pathway activation in subgroups of RMS in both human and mice. Frequently, RMS also harbor activating Ras mutations that can occur in all 3 Ras genes. Since Ras can modulate Hh signaling activity, we analyze the effects of all 3 oncogenic Ras mutations on Hh-associated tumor growth using cell lines and mouse models.

Targeting the hedgehog pathway in rhabdomyosarcoma

Children with recurrent rhabdomyosarcoma (RMS) bear a very poor prognosis, underscoring the urgent medical need to develop novel therapeutic strategies. Recently, we identified aberrant hedgehog (Hh) pathway activation in subgroups of RMS, which correlates with poor survival. The current project aims at evaluating the HH pathway as a therapeutic target in RMS as a prototypic HH-driven pediatric cancer. The experiments are performed both in cell culture and in mice.

Role of LEF1 in rhabdomyosarcoma

Rhabdomyosarcoma (RMS) develops from skeletal muscle cells that have failed to fully differentiate and is the most common soft tissue sarcoma in children and WNT signaling plays an important role in muscle development, however only few data have been published with respect to RMS. We analyze the role of LEF1, which is an important component of the WNT signaling pathway in RMS, using cell lines and mouse models.

The role of Hedgehog signaling in cutaneous squamous cell carcinoma

The role of the Hedgehog (Hh) pathway in cutaneous squamous cell carcinoma (cSCC) is far from clear. The aim of this project is to unravel the role of the Hh signaling pathway in cSCC and to investigate its potential molecular interactions with the RAS/ERK, PI3K/AKT or mTOR signaling cascades.

The role of the Hedgehog signaling cascade in the adult pituitary gland and in the development of pituitary adenoma

The Hedgehog (HH) signaling pathway plays an essential role during the development of the pituitary gland, which represents a gland with superior function in the neuroendocrine system. However the knowledge about the function of the HH signaling cascade in the adult pituitary gland and in pituitary adenoma formation is sparse. By using mouse models and human tissue samples we are investigating how HH signaling influences the development of hormone expressing cells, the hormone regulation of the gland and the development of adenoma. Additionally we are validating if Smoothened inhibitors are potential treatment options against pituitary tumors.

Adult stem cells of skin and their role in the development of skin cancer

Adult stem cells of the skin are essential for the natural regeneration of the skin and wound healing. However they also play a crucial role in the formation of skin cancer. In this project we are using mouse models and human skin samples to analyze a newly described stem cell-like cell population of the skin and its function in the normal skin homoeostasis as well as in the development of skin cancer.
